  1. Set the spectrophotometer to 410 nm and select the “Absorbance mode”.

  2. Use the blank (Tube No. 11) in both the cuvettes to set the spectrophotometer readings to ZERO.

  3. Measure the absorbance of tubes 1 – 10 against the blank and record the readings in the table 12.1.

  4. Plot the absorbance values against the PNP concentration.

  5. Fit the data points using linear regression to obtain the standard curve.
